    摘要:

    为找出从血袋内分离出外周血单个核细胞的最佳方法,同时验证在经过冻存细胞后能保有其生物活性。首先以2 516 g、4 ℃的条件下离心血袋10 min。之后以分离液分离出PBMC(757 g,离心20 min)。冻存后进行培养,可产出95%以上的毒杀功能性T细胞。研究提供了从血袋取得PBMC的方法,此方法的优势为可以一次性大量取得,减少个体状况差异,优化细胞培养过程,可大量生产不同针对性的免疫细胞,作为日后异体间配型免疫治疗使用,减少生物资源浪费,加速科研资源转化。

    Abstract:

    The purpose of the research is to find out the best method to separate the peripheral blood mononuclear cells (PBMC) from the blood bag, and verify that the mononuclear cells produced by this method can maintain their biological activity after cryopreservation.Firstly, the blood bag was centrifuged with blood bag centrifuge at 2 516g, 4 ℃ for 10 minutes. After obtaining buffy coat, PBMC were separated with blood cell separation solution with 757g for 20 minutes. In this condition, it shows better result of collection. After expansion culture from one-year cryopreservation, more than 95% of effective T cells can be obtained from collected PBMC.The study provides a method to obtain PBMC from blood bags for subsequent use. The advantages of this method are as follows: Cells can be obtained in large quantities at once to reduce individual differences and optimize cell culture process . It can produce different specific immune cells, which can be used as allogeneic immunotherapy in the future. Reduce the waste of biological resources and accelerate the transformation of scientific research resources.
